If they follow the graphic novel then yes a ton of the main characters get killed. There should be a fairly big one by seasons end.

Did Tony get killed in the first episode or the first season? No.  Sorry that's retarded and I don't need to read the books to guess that the cop does not die in season one.

And yeah.. series do kill off main characters, but they don't kill off the protagonist... and I'm not counting a Journey song.

Wild Bill Hickock was killed in the first season, but he wasn't a major character, just a big name.  And he was killed in Deadwood that year, so it was only right.
